Nov. 27, 2013 — Two months ago astronomers created a new 3D map of stars at the centre of our Galaxy (the Milky Way), showing more clearly than ever the bulge at its core. Previous explanations suggested that the stars that form the bulge are in banana-like orbits, but a paper published this week in Monthly Notices of the Royal Astronomical Society suggests that the stars probably move in peanut-shell or figure of eight-shaped orbits instead.The difference is important; astronomers develop theories of star motions to not only understand how the stars in our galaxy are moving today but also how our galaxy formed and evolves. The Milky Way is shaped like a spiral, with a region of stars at the centre known as the “bar,” because of its shape. In the middle of this region, there is a “bulge” that expands out vertically.In the new work Alice Quillen, professor of astronomy at the University of Rochester, and her collaborators created a mathematical model of what might be happening at the centre of the Milky Way. Unlike the Solar System where most of the gravitational pull comes from the Sun and is simple to model, it is much harder to describe the gravitational field near the centre of the Galaxy, where millions of stars, vast clouds of dust, and even dark matter swirl about. In this case, Quillen and her colleagues considered the forces acting on the stars in or near the bulge.As the stars go round in their orbits, they also move above or below the plane of the bar. When stars cross the plane they get a little push, like a child on a swing. At the resonance point, which is a point a certain distance from the centre of the bar, the timing of the pushes on the stars is such that this effect is strong enough to make the stars at this point move up higher above the plane. (It is like when a child on the swing has been pushed a little every time and eventually is swinging higher.) These stars are pushed out from the edge of the bulge.The resonance at this point means that stars undergo two vertical oscillations for every orbital period. …

Nov. 27, 2013 — Drugs delivered by nanoparticles hold promise for targeted treatment of many diseases, including cancer. However, the particles have to be injected into patients, which has limited their usefulness so far.Now, researchers from MIT and Brigham and Women’s Hospital (BWH) have developed a new type of nanoparticle that can be delivered orally and absorbed through the digestive tract, allowing patients to simply take a pill instead of receiving injections.In a paper appearing in the Nov. 27 online edition of Science Translational Medicine, the researchers used the particles to demonstrate oral delivery of insulin in mice, but they say the particles could be used to carry any kind of drug that can be encapsulated in a nanoparticle. The new nanoparticles are coated with antibodies that act as a key to unlock receptors found on the surfaces of cells that line the intestine, allowing the nanoparticles to break through the intestinal walls and enter the bloodstream.This type of drug delivery could be especially useful in developing new treatments for conditions such as high cholesterol or arthritis. Patients with those diseases would be much more likely to take pills regularly than to make frequent visits to a doctor’s office to receive nanoparticle injections, say the researchers.”If you were a patient and you had a choice, there’s just no question: Patients would always prefer drugs they can take orally,” says Robert Langer, the David H. Koch Institute Professor at MIT, a member of MIT’s Koch Institute for Integrative Cancer Research, and an author of the Science Translational Medicine paper.Lead authors of the paper are former MIT grad student Eric Pridgen and former BWH postdoc Frank Alexis, and the senior author is Omid Farokhzad, director of the Laboratory of Nanomedicine and Biomaterials at BWH. But what if these materials could be programmed to regenerate-themselves, replenishing the damaged or missing components, and thereby extend their lifetime and reduce the need for costly repairs?That potential is now possible according to researchers at the University of Pittsburgh Swanson School of Engineering, who have developed computational models to design a new polymer gel that would enable complex materials to regenerate themselves.Principal investigator is Anna C. Balazs, PhD, the Swanson School’s Distinguished Robert v. d. Luft Professor of chemical and petroleum engineering, and co-authors are Xin Yong, PhD, postdoctoral associate, who is the article’s lead author; Olga Kuksenok, PhD, research associate professor; and Krzysztof Matyjaszewski, PhD, J.C. Warner University Professor of Natural Sciences, department of chemistry at Carnegie Mellon University.”This is one of the holy grails of materials science,” noted Dr. Balazs. “While others have developed materials that can mend small defects, there is no published research regarding systems that can regenerate bulk sections of a severed material. Nov. 21, 2013 — A team led by UC San Francisco (UCSF) scientists has identified the disruption of a single type of cell — in a particular brain region and at a particular time in brain development — as a significant factor in the emergence of autism.The finding, reported in the Nov. 21 issue of Cell, was made with techniques developed only within the last few years and marks a turning point in autism spectrum disorders (ASDs) research.Large-scale gene-sequencing projects are revealing hundreds of autism-associated genes. Scientists have begun to leverage new methods to decipher how mutations in these disparate genes might converge to exert their effects in the developing brain.The new research focused on just nine genes, those most strongly associated with autism in recent sequencing studies, and investigated their effects using precise maps of gene expression during human brain development.Led by Jeremy Willsey, a graduate student in the laboratory of senior author Matthew W. State, MD, PhD, of UCSF, the group showed that this set of genes contributes to abnormalities in brain cells known as cortical projection neurons in the deepest layers of the developing prefrontal cortex, during the middle period of fetal development.Though a range of developmental scenarios in multiple brain regions is surely at work in ASDs, the researchers said the ability to place these specific genetic mutations in one specific set of cells — among hundreds of cell types in the brain, and at a specific point in human development — is a critical step in beginning to understand how autism comes about.”Given the small subset of autism genes we studied, I had no expectation that we would see the degree of spatiotemporal convergence that we saw,” said State, an international authority on the genetics of neurodevelopmental disorders.”This strongly suggests that, though there are hundreds of autism risk genes, the number of underlying biological mechanisms will be far fewer,” he said. “This is a very important clue to advance precision medicine for autism toward the development of personalized and targeted therapies.”Complex Genetic Architecture of ASDsASDs, which are marked by deficits in social interaction and language development, as well as by repetitive behaviors and/or restricted interests, are known to have a strong genetic component.But these disorders are exceedingly complex, with considerable variation in symptoms and severity, and little consistency in the mutations among affected individuals.Instead, with the rise of new sequencing methods over the past several years, researchers have identified many rare, non-inherited, spontaneous mutations that appear to act in combination with other genetic and non-genetic factors to cause ASDs. According to some estimates, mutations in as many as 1,000 genes could play a role in the development of these disorders.While researchers have been heartened that specific genes are now rapidly being tied to ASDs, State said the complex genetic architecture of ASDs is also proving to be challenging.”If there are 1,000 genes in the population that can contribute to risk in varying degrees and each has multiple developmental functions, it is not immediately obvious how to move forward to determine what is specifically related to autism,” State said. “Without this, it is very difficult to think about how to develop new and better medications,” he said.Focusing on Nine GenesTo begin to grapple with those questions, the researchers involved in the new study first selected as “seeds” the nine genes that have been most strongly tied to ASDs in recent sequencing research from their labs and others.Importantly, these nine genes were chosen solely because of the statistical evidence for a relationship to ASDs, not because their function was known to fit a theory of the cause of ASDs. “We asked where the leads take us, without any preconceived idea about where they should take us,” said State.The team then took advantage of BrainSpan, a digital atlas assembled by a large research consortium, including co-author Nenad Šestan, MD, PhD, and colleagues at Yale School of Medicine. Based on donated brain specimens, BrainSpan documents how and where genes are expressed in the human brain over the lifespan.The scientists, who also included Bernie Devlin, PhD, of The University of Pittsburgh School of Medicine; Kathryn Roeder, PhD, of Carnegie-Mellon University; and James Noonan, PhD, of Yale School of Medicine, used this tool to investigate when and where the nine seed genes join up with other genes in “co-expression networks” to wire up the brain or maintain its function.The resulting co-expression networks were then tested using a variety of pre-determined criteria to see whether they showed additional evidence of being related to ASDs. …

Nov. 21, 2013 — Using a new method to catch elusive “jumping genes” in the act, researchers have found two human proteins that are used by one type of DNA to replicate itself and move from place to place. The discovery, described in the Nov. 21 issue of Cell, breaks new ground, they say, in understanding the arms race between a jumping gene driven to colonize new areas of the human genome and cells working to limit the risk posed by such volatile bits of DNA.Jumping genes, more formally known as transposons or transposable elements, are DNA segments with the blueprints for proteins that help to either copy the segment or remove it, then insert it into a new place in the genome. Human genomes are littered with the remnants of ancient jumping genes, but because cells have an interest in limiting such trespasses, they have evolved ways to regulate them. Most jumping genes have mutated and can no longer move, but these “rusting hulks” are still passed down from generation to generation.One exception is a jumping gene called L1, which has been so successful that copies of it make up about 20 percent of human DNA. While many of these copies are now mutated and dormant, others are still active and thus the subject of much interest from geneticists.”Human cells have evolved ways of limiting jumping genes’ activity, since the more frequently they move, the more likely they are to disrupt an important gene and cause serious damage,” says Lixin Dai, Ph.D., a postdoctoral associate at the Johns Hopkins Institute for Basic Biomedical Sciences, who led the study. To find out more about how cells control L1 and what tricks the jumping gene uses to get around these defenses, Dai and others in the laboratory of Jef Boeke, Ph.D., first induced lab-grown human cells to make large amounts of the proteins for which L1 contains the blueprints. As expected, the two types of L1 protein joined with human proteins and genetic material called RNA to form so-called ribonucleoprotein particle complexes, which L1 uses to “jump.”To find out which human proteins interact with ribonucleoproteins — and are therefore likely to have a role in either tamping down its activity or helping it along — Boeke’s team collaborated with researchers at The Rockefeller University who had developed a technique for fast-freezing yeast with liquid nitrogen, then grinding it up for analysis with steel balls and very rapidly pulling out the ribonucleoproteins with tiny magnetic particles. “It’s a good way of preserving the interactions,” Dai says.Adapting this powerful technique to human cells, the team found 37 proteins that appear to interact with the ribonucleoprotein, and they selected two for further analysis. …

Nov. 20, 2013 — A Florida State University scientist has uncovered what may be the first recognized example of ancient Martian crust.The work of Munir Humayun — a professor in FSU’s Department of Earth, Ocean and Atmospheric Science and a researcher at the National High Magnetic Field Laboratory (MagLab) — is based on an analysis of a 4.4 billion-year-old Martian meteorite that was unearthed by Bedouin tribesmen in the Sahara desert. The rock (NWA 7533) may be the first recognized sample of ancient Martian crust and holds a wealth of information about the origin and age of the Red Planet’s crust.Humayun’s groundbreaking discoveries about the crust and what it reveals about the Red Planet’s origins will be published in the journal Nature.In order to detect minute amounts of chemicals in this meteorite, Humayun and his collaborators performed complex analysis on the meteorite using an array of highly sophisticated mass spectrometers in the MagLab’s geochemistry department. High concentrations of trace metals such as iridium, an element that indicates meteoritic bombardment, showed that this meteorite came from the elusive cratered area of Mars’ southern highlands.”This cratered terrain has been long thought to hold the keys to Mars’ birth and early childhood,” Humayun said.While craters cover more than half of Mars, this is the first meteoric sample to come from this area and the first time researchers are able to understand Mars’ early crustal growth.Using the chemical information found in pieces of soil contained in the meteorite, the researchers were able to calculate the thickness of Mars’ crust. Their calculation aligned with estimates from independent spacecraft measurements and confirms that Mars did not experience a giant impact that melted the entire planet in its early history.Using a powerful microprobe at Curtin University in Perth, Australia, the team dated special crystals within the meteorite — called zircons — at an astounding 4.4 billion years old.”This date is about 100 million years after the first dust condensed in the solar system,” Humayun said. “We now know that Mars had a crust within the first 100 million years of the start of planet building, and that Mars’ crust formed concurrently with the oldest crusts on Earth and the Moon.”Humayun and his collaborators hypothesize that these trailblazing discoveries are just the tip of the iceberg of what continued research on this unique meteorite will uncover. It saved my life. But before I underwent the stem cell transplant as part of my acute myeloid leukemia (AML) treatment, I had seriously considered backing out.


My AML treatment plan

A stem cell transplant was not what I’d been hoping for. I’d hoped that the clinical trial I was on as part of my AML treatment would put me in remission.


However, that was not to be. So my leukemia doctor at MD Anderson sent me to the stem cell unit for a transplant.


When I landed at the airport in Houston to go meet with a stem cell doctor at MD Anderson, I met a man and told him why I was there. Like many I met on my AML journey, he encouraged me and said I would be in his prayers.


Afterwards, I went to MD Anderson for final testing and an appointment with Chitra Hosing, M.D., professor in Stem Cell Transplant and Cellular Therapy. I told her my concerns about the transplant, and she reassured me. She never pressed me to go forward, but she talked about windows of opportunity. I was in remission, albeit incomplete, and remission was the time for a stem cell transplant. I left her office ready to move forward.


Second thoughts about my stem cell transplant

The next Saturday, my daughter-in-law, Ali, and I got in the car to drive to the hospital for my transplant.


About halfway there I got a call from the sister of a friend I had made at the transfusion unit. Her brother, Jesus, was a leukemia patient, like me. I had been jealous of him because he’d gotten his treatment and gone home to El Paso to recover. I had really thought that is what I would be doing, too.


Jesus’ sister told me that he died from chemo poisoning. The call could not have come at a worse time. I could no longer think about myself. This woman had just lost her brother. He’d left behind two young children and a wife.

I told her I was sorry, and as soon as I hung up, I related what had happened to Ali.


I wondered if this might be a sign. He’d died of chemo poisoning. I was about to undergo massive chemo to administer my stem cell transplant. Was I going to meet the same fate? I told Ali my concerns.


Ali dropped me off in front of the hospital. I told her not to come in because my 5-month-old grandson was asleep in the car. I then told her I would call her if I changed my mind.


Finding my resolve

I stood in front of the hospital doors not knowing what to do. A call home to my wife relieved some of my fears, but I was still apprehensive.


As I thought about it, it became clear to me that going home was not an option. I had been home for visits. Every other day was spent at the transfusion unit of the local hospital. My platelet count hovered between 10 and 15 unless I got a transfusion, and my red cell count was not much better. I turned and walked through doors with resolve.


I would check in. I would face this bravely, heeding the words of my oldest brother, my stem cell donor, who told me how I acted would impact my children.


In the morning, I asked my doctor about chemo poisoning. He just chuckled and reassured me. Two of my children came to see me that day, and I held my tears in check as I assured them that Dad was going to be okay.


And three years later, I am. I know with some certainty that I would not be here had I walked away from this life saving transplant. The stranger I had met at the airport will never know that his prayers, and the prayers of many others, for me were answered.

Researchers are using oxygen to grow large single graphene crystals on copper. Large single-crystal graphene is of great interest because the grain boundaries in polycrystalline material have defects, and eliminating such defects makes for a better material.

Nov. 14, 2013 — When it comes to the growth of graphene — an ultrathin, ultrastrong, all-carbon material — it is survival of the fittest, according to researchers at The University of Texas at Austin.The team used surface oxygen to grow centimeter-size single graphene crystals on copper. The crystals were about 10,000 times as large as the largest crystals from only four years ago. Very large single crystals have exceptional electrical properties.”The game we play is that we want nucleation (the growth of tiny ‘crystal seeds’) to occur, but we also want to harness and control how many of these tiny nuclei there are, and which will grow larger,” said Rodney S. Ruoff, professor in the Cockrell School of Engineering. “Oxygen at the right surface concentration means only a few nuclei grow, and winners can grow into very large crystals.”The team — led by postdoctoral fellow Yufeng Hao and Ruoff of the Department of Mechanical Engineering and the Materials Science and Engineering Program, along with Luigi Colombo, a material scientist with Texas Instruments — worked for three years on the graphene growth method. The team’s paper, “The Role of Surface Oxygen in the Growth of Large Single-Crystal Graphene on Copper,” is featured on the cover of the Nov. 8, 2013, issue of Science.One of the world’s strongest materials, graphene is flexible and has high electrical and thermal conductivity that makes it a promising material for flexible electronics, solar cells, batteries and high-speed transistors. The team’s understanding of how graphene growth is influenced by differing amounts of surface oxygen is a major step toward improved high-quality graphene films at industrial scale.The team’s method “is a fundamental breakthrough, which will lead to growth of high-quality and large area graphene film,” said Sanjay Banerjee, who heads the Cockrell School’s South West Academy of Nanoelectronics (SWAN). “By increasing the single-crystal domain sizes, the electronic transport properties will be dramatically improved and lead to new applications in flexible electronics.”Graphene has always been grown in a polycrystalline form, that is, it is composed of many crystals that are joined together with irregular chemical bonding at the boundaries between crystals (“grain boundaries”), something like a patch-work quilt. …

Each day in the United States, about 4,000 kids smoke their

first cigarette. Many of them will become daily smokers.


“For teens, it may seem cool to smoke. But tobacco use at a

young age can cause immediate and long-term health problems like cancer,” says Alexander Prokhorov,

M.D., Ph.D.
, director of the Tobacco Outreach Education Program at MD

Anderson.


Recent data shows that the declining number of teen

tobacco users
has stalled. And, the tobacco industry may be to blame.


The industry advertises products, like e-cigarettes,

flavored cigarillos and hookahs, as “safe” and is capturing the attention of

kids.


“All tobacco products are dangerous,” Prokhorov says. “We

need to be proactive about educating our communities, schools and governments

about the dangers of these products.”


Use the facts below to educate kids about the health risks

of trendy tobacco products.


Cigarillos contain

dangerous chemicals
Cigar

use
among high school students rose from 7% in 2009 to 12% in 2011. One

main factor: flavored cigarillos.




Cigarillos, or little cigars, are a cousin to the flavored

cigar. They’re sweet, cheap and come in colorful packages. “They’re especially

appealing to minorities,” Prokhorov says. “The tobacco industry recognizes this

and markets to them.”


You can buy cigarillos individually for less than 70 cents.


Health risks: Cigarillos

may seem fun to kids, but they contain the same dangerous chemicals as regular

cigarettes and cigars. Smoking them can cause heart disease, lung cancer and

lung disease.


Hookahs promote

nicotine addiction
Hookahs are water pipes that create flavored tobacco vapor.

Most people use hookahs at bars and cafes to socialize, but companies also sell

hookahs for personal use.


Health risks: During the average 40-minute hookah

session, users can inhale about 100 times the amount of smoke that’s in a

cigarette.


“The culprit is still nicotine and cancer-causing chemicals,”

Prokhorov says. “Nicotine addiction in youth can develop fast and remain

strong.”


E-cigarettes: a

gateway for other tobacco products
Electronic cigarettes, or e-cigarettes, are smokeless

electronic devices that the tobacco industry is pitching as harmless.


E-cigarette users inhale a vapor of liquid nicotine similar

to the way a smoker puffs on a cigarette. You can buy the liquid nicotine in a

wide variety flavors.


“Don’t be fooled,” Prokhorov says. “Because e-cigarettes

aren’t regulated by the U.S. Food and Drug Administration, we don’t know what chemicals

and additives are really in e-cigarettes, but they could be harmful.”


Health risks: Even

though they don’t use tobacco, e-cigarettes contain nicotine and can easily

become a gateway for teens to try other tobacco products. E-cigarettes also can

put kids at risk for lung cancer and heart disease.

ways to provide the best possible cancer treatment options for our patients as

we work toward our mission of Making Cancer History®. To help with this, we’ve

enlisted a cognitive computing system powered by IBM Watson.


This technology, known as Oncology Expert Advisor™ (OEA),

will soon be used by our doctors and researchers as part of our Moon Shots Program, starting

with our fight against leukemia.

Ultimately, we hope to use the OEA in all of our clinics to help our patients

regardless of their cancer type.


By pulling together and analyzing vast amounts of

information from patient and research databases, the OEA is expected to help our

care teams identify and fine-tune the best possible cancer

treatments
for our patients, while also alerting them to problems that arise

during a patient’s care. The OEA is also expected to help our researchers

advance new discoveries in our fight against cancer.


We recently spoke with Courtney DiNardo, M.D., assistant

professor in Leukemia, who’s been testing the OEA before it debuts in our leukemia

clinic. Here’s what she had to say.


What are the benefits

of using the OEA to organize and collect data about

our patients?
The OEA can extract patient information from various data sources and

synthesize all available medical records into a clear, concise and accurate

synopsis. It can analyze clinical information, medical history, as well as

leukemia-related information, such as specific genetic and molecular features,

and look at all available information in the context of published

evidence-based guidelines and available clinical

trials
.


The OEA also allows us to look at changes in a patient’s

condition over time, enabling us to learn a wealth of information within

seconds and answer complex medical questions with speed, accuracy and

confidence. This has an enormous potential to help us make the best cancer

treatment decisions for our patients and the best research decisions that can

help us make progress in our fight against cancer.


What does the use of

the OEA mean for the future of personalized medicine?


The OEA enables us to provide better, more personalized care through accurate

and evidence-based treatment recommendations based upon a specific patient’s

characteristics as well as his or her leukemia-specific characteristics.


The OEA also can help doctors identify the best cancer

treatment for a particular patient by identifying both the standard treatment options

and clinical trials for which a patient is eligible.


Additionally, by following a patient over time along with

the physician, the OEA helps minimize potential adverse events and optimize

management of the patient’s care at all times.


How – and how soon – will

the OEA change cancer treatment here at MD Anderson?
The OEA is currently in the testing and evaluation stage in MD Anderson’s Department

of Leukemia
. The actual timeline of when this will be available to our

leukemia patients and then expanded to patients with other types of cancer is

not currently known.


We’re hoping that soon, various medical providers, including

physicians, mid-level providers and research teams, will soon be able to use it

to assimilate complex and ever-expanding genomic data and published scientific research.

The OEA then integrates this information with patient data to provide insights

and helps us select the best cancer treatment for each and every patient.


What does the OEA

mean for cancer care beyond MD Anderson?
The OEA has the potential to “democratize” cancer care, meaning that any physician

with Internet access will be able to access the latest scientific knowledge and

MD Anderson’s expertise.


For physicians who aren’t leukemia experts, the OEA can

function as an expert second opinion, allowing them to access the same

knowledge and information we have here. This will enable doctors even in remote

areas to provide the highest standard of care to patients who can’t travel to

Houston for their cancer treatment.


As a doctor, what do

you consider the OEA’s greatest potential?


The ability to integrate patient level data such as additional medical history,

medications, alongside cancer genomics, as well as cancer therapy received and

responses, side

effects
, etc., will provide an unparalleled research platform that can be

used to generate questions, explore hypotheses and provide answers to critical

research questions.


For example, the OEA will also enable us to analyze why

certain patients respond to certain cancer treatments and why particular patients

experience certain side effects, using a research platform that is currently not

available. This gives us the potential to really learn from the vast amount of

information we currently have, and use it to advance cancer research and to benefit

each individual patient.


I’m excited about the possibilities for accelerating new

research and providing the most effective treatments and cures for our patients.

Nov. 7, 2013 — In a new study, Dartmouth researchers rule out a controversial theory that the accelerating expansion of the universe is an illusion.While the findings don’t explain the cosmic speed-up, they eliminate one provocative possibility that our planet, solar system and galaxy are at the center of the universe and that there is no dark energy. The findings appear in the journal Physical Review D.The 2011 Nobel Prize was awarded for the discovery that the expansion of the universe is accelerating. One leading idea to explain the acceleration is a new, mysterious substance called dark energy, which is thought to make up nearly three-fourths of the energy of the universe. But another alternative is that Earth, our solar system and Milky Way galaxy are at the center of the universe. That theory violates the standard assumption that the universe has no center, but if true, then cosmic acceleration could be explained without dark energy or any new laws of physics.But Dartmouth researchers found that this model can’t hold up to other observational tests. The sky glows with light left over from the Big Bang, also known as the Cosmic Microwave Background, so they calculated how that glow would be affected. Their findings show that the model’s prediction is completely contrary to the glow that has been measured.”Essentially, we held a mirror up to the universe and asked if the reflection was special,” says Robert Caldwell, a professor of physics and astronomy who co-wrote the article with undergraduate physics major Nina Maksimova. “The reflection shows that we do not appear to live in a special location, and decisively excludes this explanation for the universe’s accelerating expansion. It would be a great relief to be able to understand a basic problem of cosmology within the known laws of physics, but our research is an important step in explaining the physics responsible for the cosmic acceleration.”

Skeletal muscle has proved to be very difficult to grow in patients with muscular dystrophy and other disorders that degrade and weaken muscle. Researchers now report boosting muscle mass and reversing disease in a mouse model of Duchenne muscular dystrophy, using a “cocktail” of three compounds identified through a new rapid culture system. Adding the same compounds to stem cells derived from patients’ skin cells, they then successfully grew human muscle cells in a dish.

Nov. 7, 2013 — Skeletal muscle has proved to be very difficult to grow in patients with muscular dystrophy and other disorders that degrade and weaken muscle. Researchers at Boston Children’s Hospital’s Stem Cell Program now report boosting muscle mass and reversing disease in a mouse model of Duchenne muscular dystrophy, using a “cocktail” of three compounds identified through a new rapid culture system. Adding the same compounds to stem cells derived from patients’ skin cells, they then successfully grew human muscle cells in a dish.Senior investigator Leonard Zon, MD, director of the Stem Cell Program at Boston Children’s and an investigator with the Howard Hughes Medical Institute, hopes for clinical trials to begin transplanting these cells into patients with muscle loss in the next several years. In the meantime, the screening technique, described in the journal Cell (published online November 7), can also be applied to multiple disorders beyond muscular dystrophy, he says.#1: ZebrafishThe drug screening technique begins with zebrafish. Zon and colleagues placed tiny zebrafish blastomeres — cells from very early stage embryos — into 384-well culture dishes, adding one test compound to each well. This allowed them to screen 2,400 compounds at once, seeking ones that boosted numbers of muscle progenitor cells in the see-through fish as they developed.The Zon Lab’s earlier work with zebrafish led to a drug derived from prostaglandin that expands numbers of blood stem cells and is now in a Phase II clinical trial. But this time, researchers genetically modified the fish so that newly created muscle progenitor cells would show up in fluorescent green, and muscle fibers in red. This enabled the team to rapidly quantify muscle cells and fibers produced under different conditions, based on visual readouts — without having to examine each well’s contents through laborious biochemical techniques.”Automated imagers can take a picture to assay all the 384 wells within a matter of a week, as compared with several months with our earlier technique,” says Zon, who is also a member of Boston Children’s Division of Hematology/Oncology as well as the Harvard Stem Cell Institute (HSCI).#2: MiceThe zebrafish tests revealed six compounds that expanded numbers of skeletal muscle progenitor cells.
